Our group's doctoral student Nan Li won the poster award awarded by Royal Society of Chemistry, Analyst Journal, with the title of "A Peptide Conjugation Membrane Type-1 Matrix Metalloproteinase Responsive Probe for Real-Time Visualization of Cell Migration on a Microfluidic Chip". This study fabricated a three-channel microfluidic chip to simulate the cell-culture microenvironment that dually regulates biochemical factors and shear stimulation. At the same time, an enzyme-activated probe was designed to indicate the activity of cell-surface protein. This probe is covalently bound to the cell by a crosslinker. The distance between the FRET pair was within the range of 1–10 nm, so that the donor signal could be quenched by the acceptor. While, in the presence of MT1-MMP, the indicator was cleaved from the active site, leading to the donor fluorescence recovery. This method provides a feasible idea for the study of specific proteins on the cell membrane.
